Yael Tayeb at Cambridge has contributed to the biology of ATR kinase and the rationale for ATR inhibition in cancer therapy. The ATR inhibitor ceralasertib and others have entered clinical trials based on the principle that cancer cells with oncogene-driven replication stress are particularly dependent on ATR to prevent replication fork catastrophe — an exploitable synthetic lethality opportunity in RAS-mutant and other oncogene-activated cancers.
