William Sellers at the Broad Institute has contributed to allosteric kinase inhibitor development and the systematic discovery of cancer dependencies through large-scale functional genomic approaches including the Cancer Dependency Map. His research on PI3K pathway inhibitors contributed to understanding the selectivity challenges and therapeutic opportunities of allosteric PI3K inhibition. He has been involved in development of approaches that leverage allosteric mechanisms to achieve greater selectivity over ATP-competitive inhibitors. His functional genomics work on kinase dependencies has revealed synthetic lethal interactions that enable combination kinase inhibitor strategies.
