Robert Weinberg demonstrated the critical connection between epithelial-mesenchymal transition (EMT) and cancer stem cell properties, showing that Wnt and Notch signaling drive EMT programs that confer stem cell-like self-renewal, drug resistance, and tumor-initiating capacity on differentiated cancer cells. His discovery that EMT-generated cancer stem cells exhibit plasticity between stem and non-stem states through Wnt-dependent mechanisms revealed that cancer stem cell identity is dynamic rather than fixed. His foundational work on oncogenes and tumor suppressor genes provided the framework for understanding how developmental pathway mutations drive cancer initiation.
