Stephen Elledge at Harvard has made foundational contributions to understanding the DNA damage response (DDR) — the network of cell cycle checkpoint, repair, and apoptosis signals activated by DNA damage. His discoveries of the ATR-CHK1 and ATM-CHK2 checkpoint kinase cascades, of the role of the DDR in suppressing oncogenic transformation, and of the concept that replication stress is an early activator of the DDR in premalignant cells have been highly influential in cancer biology and therapy.
