Shimon Sakaguchi at Osaka University discovered regulatory T cells (Tregs) marked by Foxp3 expression — a discovery that fundamentally changed immunology by explaining how the immune system maintains self-tolerance and how cancer exploits Treg-mediated immunosuppression. His work demonstrating that tumor-infiltrating Tregs suppress anti-tumor immunity provided the rationale for anti-CTLA-4 therapy's mechanism of depleting intratumoral Tregs, and for Treg-depleting strategies as cancer immunotherapy.
