Scott Kern identified SMAD4 (DPC4) as a tumor suppressor gene deleted in pancreatic cancer, establishing the first direct genetic link between TGF-beta pathway loss and one of the deadliest human cancers. His discovery that SMAD4 is inactivated in approximately 55% of pancreatic ductal adenocarcinomas revealed that loss of TGF-beta growth inhibitory signaling is a hallmark of pancreatic cancer progression. He showed that SMAD4 loss correlates with widespread metastatic disease at diagnosis, identifying SMAD4 as a biomarker for metastatic potential. His work defined the genetic basis for TGF-beta pathway disruption in pancreatic cancer and its clinical implications.
