Ze'ev Ronai has elucidated molecular mechanisms underlying TTFields anti-tumor activity, demonstrating that alternating electric fields disrupt the unfolded protein response and endoplasmic reticulum stress pathways, leading to proteotoxic crisis and autophagy-dependent cell death in cancer cells. His research revealed that TTFields alter protein aggregation dynamics and chaperone function, creating synthetic lethality with proteasome inhibitors. His work on stress-responsive ubiquitin ligases provided mechanistic insights into why rapidly dividing cancer cells are preferentially affected by TTFields.
