Roberta Zappasodi is a tumor immunologist who has made fundamental contributions to understanding how CTLA-4 blockade depletes intratumoral regulatory T cells via Fc-receptor-dependent mechanisms, which may explain the differential activity of ipilimumab versus tremelimumab. Her work at Weill Cornell dissects the relative contributions of cell-intrinsic CTLA-4 checkpoint function and Fc-mediated effector mechanisms in combination immunotherapy. These findings have implications for the engineering of next-generation CTLA-4 antibodies with superior tumor-selective Treg depletion and reduced toxicity.
