Robert Weinberg is a co-discoverer of the first human oncogene (RAS) and the first tumor suppressor gene cloned (RB1), and co-authored the landmark Cancer Hallmarks framework with Douglas Hanahan. He has defined the molecular basis of oncogenesis, tumor suppression, and EMT-driven cancer invasion and metastasis over five decades.

First Human Oncogene Isolation (RAS)

Isolated and characterized the first transforming oncogene from a human bladder carcinoma cell line as a mutated version of the normal HRAS proto-oncogene, defining oncogene activation by point mutation.

RB1 Cloning — First Tumor Suppressor Gene

Cloned RB1, the retinoblastoma susceptibility gene, as the first tumor suppressor gene, establishing the two-hit hypothesis molecularly and defining a new class of cancer genes controlling cell cycle entry.

Epithelial-Mesenchymal Transition (EMT) in Metastasis

Demonstrated that EMT programs enable carcinoma cells to acquire invasive and stem-cell properties, and that mesenchymal-epithelial transition (MET) facilitates metastatic colonization, providing a mechanistic basis for metastasis.
