Robert Schreiber formulated the cancer immunoediting hypothesis — the unifying framework of elimination, equilibrium, and escape that describes how the immune system shapes cancer evolution — providing the conceptual foundation for understanding why immunotherapy works and why it fails. His experimental demonstration that the immune system eliminates nascent cancers, holds surviving cancer cells in equilibrium, and eventually selects for immune-escape variants that form clinically apparent tumors revolutionized tumor immunology. He provided definitive experimental proof for cancer immunosurveillance using IFN-gamma receptor knockout and RAG knockout mice. His conceptual framework guides rational immunotherapy design and combination strategies.
