Robert Abraham has made seminal contributions to understanding ATM and ATR checkpoint kinase signaling in the DNA damage response and its relevance to cancer. His research characterized how ATR responds to replication stress and stalled forks to activate cell cycle arrest and DNA repair, while ATM responds primarily to double-strand breaks. He identified key substrates and effectors of ATR/ATM including Chk1 and Chk2 that transmit checkpoint signals and coordinate with CDK inhibitory phosphorylation to arrest proliferation. His work bridged basic checkpoint biology with cancer therapeutic opportunities including ATR inhibitors, Chk1 inhibitors, and WEE1 inhibitors now in clinical development.
