Richard Young defined the concept of super-enhancers -- large clusters of enhancers densely loaded with Mediator, BRD4, and master transcription factors that drive expression of cell identity genes and key oncogenes in cancer. He demonstrated that cancer cells acquire super-enhancers at oncogenes through various mechanisms including focal amplification, chromosomal translocation, and transcription factor overexpression, creating exceptional dependencies on transcriptional coactivators. His laboratory proposed that super-enhancers function through phase separation, forming biomolecular condensates that concentrate transcriptional machinery. His super-enhancer concept has become a major organizing framework for understanding transcriptional dysregulation in cancer.
