Richard Simon at NCI developed widely-used statistical methods for cancer clinical trials, including the Simon two-stage optimal design for Phase II trials and methodology for biomarker-enrichment designs (testing targeted therapies in molecularly-defined patient subsets). His frameworks for validating predictive biomarkers and designing biomarker-driven trials have been foundational for the development of precision oncology therapeutics.
