Richard Fishel at Ohio State made key contributions to understanding the molecular mechanism of DNA mismatch repair (MMR) — identifying the MutS homologue MSH2 and characterizing how MMR proteins recognize and correct replication errors. His work elucidating the MMR reaction mechanism and demonstrating that MMR deficiency causes microsatellite instability and Lynch syndrome has been central to understanding this cancer predisposition syndrome and to the development of immunotherapy for MMR-deficient cancers.
