Randal Kaufman has made foundational contributions to understanding the unfolded protein response (UPR) — the cellular stress response to endoplasmic reticulum (ER) overload. His work on the IRE1, PERK, and ATF6 signaling branches of the UPR and how cancer cells exploit UPR signaling for survival under proteotoxic stress is particularly relevant to myeloma, in which high immunoglobulin production creates constitutive ER stress managed by UPR upregulation.
