Raul Rabadan applied computational genomics to map the mutational landscape of Wnt, Notch, and Hedgehog pathway components across cancer types, revealing that 93 percent of colorectal cancers harbor mutations activating Wnt signaling and identifying synthetic lethal interactions between pathway components exploitable for cancer stem cell targeting. His tumor phylogenetic analyses demonstrated that cancer stem cell populations evolve through branching clonal architectures with pathway-specific selection pressures. He develops machine learning models predicting cancer stem cell vulnerability to pathway-targeted therapies based on multi-omic tumor profiling.
