Philip Beachy co-discovered the Hedgehog signaling pathway in mammals and demonstrated its critical role in cancer, particularly basal cell carcinoma and medulloblastoma, where constitutive pathway activation drives tumor growth. His laboratory elucidated the mechanism by which Patched and Smoothened transduce Hedgehog signals, directly enabling the development of vismodegib and sonidegib as the first FDA-approved Hedgehog pathway inhibitors for advanced BCC. He showed that paracrine Hedgehog signaling from tumor cells to stromal fibroblasts creates a supportive tumor microenvironment in pancreatic and other cancers. His foundational work established developmental signaling pathways as tractable cancer targets.
