Philip Beachy co-discovered the Hedgehog signaling pathway and characterized its critical role in development and cancer. His laboratory identified the Smoothened (SMO) receptor and Patched (PTCH1) tumor suppressor as core components of the pathway. He demonstrated that activating SMO mutations and PTCH1 loss drive basal cell carcinoma, motivating the development of SMO inhibitors including vismodegib. His foundational discoveries enabled the first targeted therapy for basal cell carcinoma and opened new avenues for treating other Hedgehog-driven cancers.
