Peter Cresswell has defined the molecular machinery of MHC class I antigen presentation, discovering the peptide loading complex containing TAP, tapasin, calreticulin, and ERp57. His foundational work established how peptides generated by the proteasome are transported into the ER and loaded onto MHC-I molecules for T cell recognition. These discoveries have been essential for understanding how cancer cells evade immune detection by disrupting antigen processing.
