Peter Adams discovered senescence-associated heterochromatic foci (SAHF), demonstrating that senescent cells undergo dramatic chromatin reorganization that stably silences E2F target genes required for cell proliferation. His work showed that the histone chaperone HIRA and HP1 proteins drive SAHF formation, creating a robust epigenetic barrier to cell cycle re-entry. He elucidated how SAHF contribute to the irreversibility of senescence and how their disruption can lead to senescence bypass and cancer progression. His research on chromatin dynamics in senescence has informed the development of epigenetic approaches to reinforce or exploit senescence in cancer therapy.
