Jonathan Ostrem co-discovered the covalent binding approach to KRAS G12C that enabled the entire class of clinical KRAS inhibitors. His structural biology work at UCSF defined the switch II pocket that sotorasib and adagrasib exploit. This foundational discovery transformed KRAS from an undruggable to a treatable oncogenic target.
