Roel Nusse co-discovered the Wnt gene family as proto-oncogenes activated by mouse mammary tumor virus integration, founding the entire Wnt signaling field and its central role in cancer stem cell biology. His biochemical characterization of Wnt ligand lipid modification, Frizzled receptor binding, and LRP5/6 co-receptor activation defined the molecular events initiating canonical Wnt signaling that maintains cancer stem cell self-renewal. His demonstration that purified Wnt3a sustains intestinal stem cell growth in culture provided the foundation for cancer organoid technology and Wnt-targeted therapy development.
