Nicole Gaudelli invented the adenine base editor (ABE), which converts A-to-G (and T-to-C) base pairs with high precision, and has translated base editing technology toward therapeutic applications in cancer. Her work at Beam Therapeutics has advanced multiplexed base editing for engineering enhanced CAR-T cells with simultaneous knockout of T cell exhaustion genes, HLA genes for allogeneic compatibility, and insertion of novel receptor constructs. She has characterized the safety profile of base editors including off-target editing and demonstrated their therapeutic potential exceeds traditional CRISPR nucleases for precision cancer cell engineering. Her work is driving the clinical development of base-edited cell therapies for hematologic malignancies.
