Neville Sanjana has advanced CRISPR screen library design and application, developing optimized genome-scale and targeted libraries for identifying cancer vulnerabilities in both coding and non-coding regions of the genome. His laboratory pioneered CRISPR tiling screens across enhancers and regulatory elements to identify non-coding sequences essential for oncogene expression, revealing that cancer dependencies extend beyond protein-coding genes. He has conducted genome-wide CRISPR screens to identify mechanisms of cancer immune evasion, discovering genes whose loss enables tumor cells to escape T cell-mediated killing. His work on immunotherapy resistance screens has identified new targets for combination therapy strategies.
