Alessandro Moretta discovered the family of natural cytotoxicity receptors (NCRs) including NKp30, NKp44, and NKp46, which are the principal activating receptors responsible for NK cell-mediated tumor killing, fundamentally transforming understanding of innate anti-tumor immunity. His demonstration that KIR-ligand mismatch in haploidentical stem cell transplantation produces potent graft-versus-leukemia effects without graft-versus-host disease revolutionized transplant immunology. His pioneering contributions to NK cell biology created the conceptual foundation for the entire NK cell therapy field.
