Matthew Macauley is a leader in understanding how tumor sialic acid glycans engage Siglec inhibitory receptors on immune cells to create a glyco-immune checkpoint that suppresses anti-tumor immunity, and is developing strategies to block this interaction therapeutically. His work demonstrated that Siglec-7 and Siglec-9 on NK cells and macrophages are inhibited by tumor sialoglycans, providing a mechanism for innate immune evasion that is independent of classical immune checkpoints. He has developed sialic acid-based ligands and anti-Siglec antibodies that enhance anti-tumor immune responses. His research establishes the Siglec-sialoglycan axis as a major immunotherapeutic target.
