Mark Greene contributed to early characterization of the HER2/neu oncogene and its role in breast cancer, including developing anti-HER2 antibodies in the preclinical context that contributed to the therapeutic rationale for antibody-based HER2 targeting. His laboratory studied the structural basis of HER2 signaling and approaches to antibody-mediated receptor interference. His foundational work on HER2 antibody binding mechanisms has informed the development of not only trastuzumab but subsequent generations of anti-HER2 therapeutics including bispecific antibodies and antibody-drug conjugates.
