Mark Davis at Caltech developed cyclodextrin-based polymer nanoparticles (RONDEL system) for siRNA delivery and led the first human demonstration of RNAi mechanism of action — showing in patients that systemically delivered siRNA nanoparticles reached tumor tissue and silenced target gene expression through RNAi. This proof-of-concept clinical demonstration was a landmark for nanomedicine-enabled nucleic acid therapeutics.
