Manolis Bhatt has been a key contributor to understanding how necroptotic cell death drives immunogenic responses in cancer, demonstrating that RIPK3-dependent necroptosis is superior to apoptosis in promoting dendritic cell cross-presentation and CD8+ T cell priming against tumor antigens. His research showed that necroptotic cancer cells release HMGB1, ATP, and calreticulin in patterns distinct from apoptotic cells, engaging different innate immune receptors. He developed necroptosis-based cancer vaccines using RIPK3-activated tumor cell lysates that provide durable anti-tumor immunity in preclinical models. His work has established necroptosis induction as a strategy to enhance cancer immunogenicity.
