Lucio Miele

Lucio Miele has been a leading clinical investigator of Notch pathway inhibition in solid tumors, demonstrating that NOTCH2 and NOTCH3 drive cancer stem cell maintenance in triple-negative breast cancer and other aggressive solid tumor subtypes. His research established that Notch signaling promotes epithelial-mesenchymal transition, chemotherapy resistance, and tumor-initiating cell survival in breast cancer. He led clinical trials of gamma-secretase inhibitors in combination with chemotherapy showing preferential activity in Notch-high breast cancer subtypes. His work on DLL4-Notch signaling in tumor angiogenesis has identified novel anti-angiogenic strategies beyond VEGF targeting.
