Luca Pellegrini has elucidated the structural basis of how cancer-associated mutations in U2AF1 and SRSF2 alter RNA recognition and splicing fidelity, providing critical insights for drug design targeting the mutant spliceosome. His crystallographic and cryo-EM studies revealed how point mutations in these splicing factors change their RNA-binding specificity to cause genome-wide mis-splicing. He has contributed structural insights guiding the development of spliceosome modulators for cancer therapy. His structural biology work bridges the gap between cancer genetics and therapeutic intervention.
