Keiran Bhatt at Moffitt has contributed to understanding BRAF inhibitor resistance mechanisms in melanoma. BRAF V600E/K mutations are present in approximately 50% of melanomas, and while BRAF plus MEK inhibitors produce high response rates, resistance invariably develops. Research on the molecular mechanisms of acquired resistance — including NRAS mutations, MEK amplification, COT1/MAP3K8 activation, and phenotype switching — informs combination strategies to overcome resistance.
