Kai Wucherpfennig has used genome-wide CRISPR screens to systematically identify genes essential for cancer immune evasion, discovering that IFN-gamma pathway components and MHC-I regulators are critical determinants of immunotherapy response. His identification of CMTM6 as a key regulator of PD-L1 surface stability opened new therapeutic approaches. He has revealed how cancer cells exploit defects in IFN-gamma signaling to escape both T cell and NK cell surveillance.
