Joseph Burchenal was a pioneering chemotherapy investigator at Memorial Hospital, the forerunner of Memorial Sloan Kettering Cancer Center, who conducted early clinical investigations of 6-mercaptopurine, methotrexate, and other antimetabolites in childhood and adult leukemia during the 1940s-1960s. His clinical research helped establish the therapeutic activity of 6-MP in acute leukemia following its chemical synthesis by George Hitchings and Gertrude Elion, contributing to the first sustained remissions in leukemia patients. He was instrumental in building Memorial Hospital's chemotherapy program into one of the world's leading cancer chemotherapy research units, establishing a model for dedicated oncology clinical pharmacology that influenced cancer centers globally. His early clinical chemotherapy investigations at Memorial formed a cornerstone of the emerging field of medical oncology and demonstrated that pharmacological approaches could produce meaningful clinical benefit in cancer.
