Jon Aster is a cancer pathologist whose mechanistic research has profoundly advanced understanding of NOTCH signaling in cancer. His studies characterized the activation mechanism of NOTCH1 and defined how gain-of-function mutations found in T-ALL constitutively activate the pathway. He has identified NOTCH pathway activation in multiple lymphoma subtypes beyond T-ALL and characterized downstream transcriptional targets. His work on NOTCH proteolytic processing has informed the development of therapeutic strategies to block pathological NOTCH activation.
