E. John Wherry at Penn has made major contributions to the molecular and epigenetic mechanisms of T cell exhaustion, particularly in defining the distinct epigenetic programs that distinguish exhausted T cells from functional effector and memory T cells. His work has revealed that anti-PD-1 therapy partially restores exhausted T cell function without erasing the exhaustion epigenome — explaining both the efficacy and the incomplete reinvigoration achieved with checkpoint blockade.
