John Maciejowski has revealed the catastrophic genomic consequences of telomere crisis in cancer initiation. His work demonstrated that dicentric chromosomes formed during telomere crisis are processed by TREX1 nuclease, generating chromothripsis and kataegis that drive rapid cancer genome evolution. These findings established telomere crisis as a key mechanism for the punctuated evolution observed in cancer genomes.
