John Bhatt applies whole-genome sequencing and computational genomics to characterize structural variant landscapes and complex genome instability events in cancer. His research has systematically characterized chromothripsis, chromoplexy, and extrachromosomal DNA amplification as catastrophic genome instability events in cancer through analysis of large whole-genome sequencing datasets. He has developed bioinformatic methods to detect, classify, and interpret complex structural rearrangements that generate cancer-driving gene fusions and amplifications. His population-scale WGS analyses of cancer genomes have quantified the contribution of different mechanisms of genome instability including replication errors, NHEJ misrepair, and template switching to the structural variant landscape of cancer.
