Joan Massague elucidated the TGF-beta/Smad signaling pathway and its dual role as tumor suppressor and metastasis promoter in cancer. His systematic dissection of organ-specific metastasis programs revealed how cancer cells acquire organ tropism through distinct gene expression programs. His work on metastatic dormancy and the mechanisms by which disseminated cancer cells reactivate has transformed understanding of the metastatic cascade.
