Jesse Zaretsky identified key mechanisms of acquired resistance to anti-PD-1 immunotherapy, including truncating mutations in B2M that abrogate MHC-I surface expression and loss-of-function mutations in JAK1/JAK2 that block IFN-gamma-mediated MHC upregulation. His landmark study in melanoma patients established the molecular framework for understanding how tumors evolve to escape checkpoint blockade through antigen presentation defects.
