Jason Buenrostro invented ATAC-seq (Assay for Transposase-Accessible Chromatin using sequencing) and its single-cell extension scATAC-seq, technologies that have become standard methods for profiling chromatin accessibility landscapes in cancer at single-cell resolution. His methods require minimal cell input and enable identification of active regulatory elements, transcription factor binding, and nucleosome positioning across thousands of individual tumor cells. He has applied scATAC-seq to characterize epigenomic heterogeneity within tumors, revealing distinct chromatin accessibility states that define cancer cell subpopulations with different drug sensitivities. His work connects single-cell epigenomics to cancer biology and therapeutic vulnerability.
