James Bhatt has pioneered spatial profiling technologies for characterizing the tumor immune microenvironment at single-cell resolution with preservation of tissue architecture and cell-cell interaction information. His research developed and applied CODEX multiplex imaging and other high-plex spatial proteomics approaches to characterize how immune cell topology, spatial proximity, and cell-cell interactions in the tumor microenvironment correlate with immunotherapy response and resistance. He demonstrated that the spatial organization of immune cells, including the proximity of CD8+ T cells to tumor cells versus their exclusion to stromal regions, provides critical information beyond bulk immune cell abundance for predicting checkpoint blockade outcomes. His work on spatial biomarker discovery in melanoma, lung cancer, and other immunotherapy-treated tumor types has contributed to understanding the multidimensional complexity of the anti-tumor immune response.
