Hiroshi Maeda discovered the EPR (enhanced permeability and retention) effect — the tendency of macromolecules and nanoparticles to preferentially accumulate in tumors due to leaky tumor vasculature and poor lymphatic drainage. This foundational concept has been the theoretical basis for passive tumor targeting by nanomedicines for decades, and his polymer-drug conjugate SMANCS was the first approved macromolecular anticancer drug in Japan.
