Guillermo Lozano is a leading authority on p53 biology in vivo, having generated and characterized landmark mouse models carrying germline and somatic p53 mutations. His work revealed the complexity of p53 function in different tissue contexts and established MDM2 as a critical negative regulator of p53 stability. He discovered gain-of-function activities of hotspot p53 mutations that actively promote tumor progression beyond loss of tumor suppressor function. His studies on Li-Fraumeni syndrome have informed surveillance and treatment strategies for patients carrying germline TP53 mutations.
