Genevieve Almouzni has made milestone contributions to understanding histone chaperone-mediated chromatin assembly and its dysregulation in cancer, particularly through her studies of CAF-1 (chromatin assembly factor 1) and the DAXX/ATRX complex. She demonstrated that CAF-1 is essential for coupling DNA replication with chromatin assembly, maintaining epigenetic marks through cell division, and that CAF-1 overexpression in cancer promotes proliferation and correlates with poor prognosis. Her work on the DAXX/ATRX histone H3.3 chaperone complex revealed how DAXX/ATRX mutations in pediatric glioblastoma and pancreatic neuroendocrine tumors cause alternative lengthening of telomeres and chromatin instability. Her research connects histone dynamics to cancer epigenome maintenance.
