Gad Asher has revealed fundamental connections between circadian rhythms, cellular metabolism, and cancer through his discovery that NAD+ levels oscillate with circadian periodicity, linking the molecular clock to SIRT1-mediated metabolic regulation relevant to tumor metabolism. His laboratory demonstrated that oxygen serves as a circadian zeitgeber capable of resetting peripheral clocks, with implications for hypoxic tumor microenvironments. He showed that disruption of circadian metabolic oscillations alters the Warburg effect and oxidative phosphorylation balance in cancer cells. His research has identified metabolic vulnerabilities in circadian-disrupted cancers amenable to chronotherapy.
