Elaine Fuchs identified the stem cells of the skin and hair follicle and demonstrated how their niche signals including Wnt, BMP, and Notch are hijacked during squamous cell carcinoma formation, establishing the paradigm that tissue stem cells are the cells of origin for epithelial cancers. Her lineage tracing experiments showed that cancer stem cells in skin tumors occupy specific microanatomical niches that provide Wnt ligands essential for maintaining their self-renewal capacity and tumor-propagating ability. Her work on stem cell competition and clonal dynamics revealed how mutant stem cells outcompete normal neighbors during cancer initiation through altered niche signaling.
