Franklin Huang discovered the mechanism by which TERT promoter mutations (C228T and C250T) activate telomerase expression in cancer. His work revealed that these mutations create de novo binding sites for the GABP transcription factor, explaining how the most common non-coding mutations in cancer reactivate telomerase. These findings opened new therapeutic strategies targeting TERT promoter-driven transcription in glioblastoma and melanoma.
