E. John Wherry pioneered the molecular characterization of CD8 T cell exhaustion and its epigenetic basis. His work defined TOX as a master regulator of the exhausted state and revealed limits to PD-1 reinvigoration. He continues to translate exhaustion biology into next-generation immunotherapies.
