Donald Berry at MD Anderson pioneered Bayesian adaptive clinical trial design in cancer, developing the I-SPY series of platform trials for early breast cancer — an innovative design that uses adaptive randomization based on biomarker subtypes to rapidly identify which experimental regimens work in which subtypes. The I-SPY framework has since been widely adopted and has efficiently identified multiple promising regimens, demonstrating the value of Bayesian adaptive design in oncology.
